TG Therapeutics (TXTG) - my Core Marketocracy holding

My responses in an article format -

TG Therapeutics (TGTX) is a major holding of my Marketocracy Portfolio and it is also one of my best stock ideas.

TGTX saw a huge 300% appreciation in 2014 and I think it will continue it sharp price move in the coming 12 months. If it isn't acquired by AbbVie before then.

TGTX’s success comes on the back of pre-clinical and clinical positive results of its two promising immunotherapy drugs TG-1101, TGR-1202, and combination therapies for treating various blood cancers. Treatment efficiency and safety profile data presented so far has impressed investors and offers blood cancer patients improved treatments with less harsh side-affects.

Their drug TG-1101 is a third generation anti-CD20 monoclonal antibody (attacks cancer cells from the outside) while their drug TGR-1202 is a next generation PI3K delta inhibitor (attacks cancer cells from the inside). The drugs can be used on their own as mono-therapies or together in combination therapy. 

In combination therapy the two drugs are a nice complement, they use a double-pronged mechanism of action to attack the same cancer cells. One drug attacking the outside of cancer cells while the other drug attacks the inside of cancer cells. These two drugs have so far show excellent safety profiles so they would also be suited for combination use with other companies antibodies and checkpoint inhibitors.

Most FDA approved antibodies and checkpoint inhibitors come with Adverse Events (AE) Label Warnings. High toxicity levels are present in same-class drugs like Roche's Rituxan and Gilead's Idelalisib. TGTX's drugs are showing much lower toxicity levels which at this stage would not require them to carry a label warning. Their drugs' safety profiles look to be best in class right now which offers TGTX a significant competitive advantage. Efficiency for these drugs looks impressive from Phase 2 trial results.       

TGTX is a small company with a MC of less than 800M. It has so far flown under-the-radar on Wall Street. I think the next 12 months are set-up for investors to really take notice of this company's potential.

This will be a busy year for TGTX, they will be rolling-out at least another two Phase 3 clinical trials and together with their ongoing clinical trails and several new combo trials to be initiated they will be generating much more investor interest. Coupled with more positive trial result announcements TGTX can expect to maintain its current share-price momentum. 


Lead Program

Enrollments have begun in TGTX's lead program. This is a Phase3 combination trial with their drug TG-1101 plus Pharmacyclics*** (PCYC) inhibitor drug Imbruvica for treating relapsed high-risk CLL. This combo study will be run against Imbruvica as a mono-therapy. Enrollment completion will be around Q3 2016. 

According to the Nation Cancer Institute we have around 100,000 CLL patients in the US and around 15,000 new CLL cases are diagnosed every year.

GlobalData forecasts that drugs Imbruvica and Idelalisib will have a total combined sales of $2B in 2018, a 60% share of total CLL market.


This Phase3 study has a high likelihood of success based on results seen from their Phase2 clinical trial. TGTX's combination demonstrated a superior patient treatment efficiency. 

Impruvica as a mono-therapy has an Overall Response Rate (ORR) near 60% while TGTX's combo showed a higher ORR of 87% . In high risk CLL patients an even higher 95% ORR (19 of 20 patients) was achieved. The Phase3 study is designed for this high risk CLL patient population.

Should this Phase3 trial achieve similar highly efficient results as seen in Phase2 TGTX will also likely win Breakthrough Therapy Designation which will be another share price catalyst.


***Two weeks ago AbbVie made a $21B acquisition of Pharmacyclics and their drug Imbruvica. This is an unexpected positive catalyst for TGTX because much of their planned treatments are in combinations with Imbruvica. TGTX, with a relatively small $800M MC may also be on AbbVie's Acquisition list.

There are several immunotherapy drugs already approved for treating CLL; like Roche's Rituxan and Gazyva, GlaxoSmithKline's Arzerra and now AbbVie's Imbruvica.

Rituxan was the dominant treatment in this space. In 2013, it generated around 7.0B in sales, which inludes CLL and other cancer indications. Roche has produced a next generation Rituxan drug, it's called Gazyva. Gazyva is proven to have a superior efficiency to Rituxan. Patients taking Gazyva plus chemo live a median 26.7 months without cancer progression (PFS) compared with 15.2 months for Rituxan plus chemo.

TGTX's CLL combo treatment so far appears superior to Roche's Gazyva in both efficiency and safety profile. Gazyva has a 76% ORR vs TGTX's 87% ORR. Gazyva carries an AE warning label while TG-1101 is unlikely to carry one.

Should TGTX reproduce similar efficiency and safety profile data in their Phase3 trial their superior treatment should be able to capture a size-able slice of this CLL immunotherapy market.


Encouraging Recent developments

TGTX reported updated data at ASH (Dec 2014) from 3 clinical trials in various Lymphomas such as Chronic Lymphocytic Leukemia (CLL), Non-Hodgkin's Lymphoma (NHL), Mantle Cell Lymphoma (MCL) and Small Lymphocytic Lymphoma (SLL).
The trial results presented demonstrated impressive treatment efficiencies alongside excellent safety profiles. The data was encouraging enough to push TGTX’s share price up over 30% intra-day.

In Sept 2014, the FDA granted SPA (Special Protocol Assessment) for TGTX's combo TG-1101 plus Imbruvica.

I have confidence TGTX's management can continue to execute their detailed plans as they demonstrated in 2014. And should their upcoming clinical results be nearly as good as those in 2014 this stock will be at least a double in a years time. That is if it's not acquired before then.

Northwest Biotherpeutics (NWBO) - offering exciting cancer vaccines

I would like to nominate NWBO as one of my best stock ideas.

NWBO is a small biotech company in the Immuno-Oncology space. They have two products in clinical trials for the treatment of solid cancers.
DCVax-L is their lead product, it’s at an advanced Phase 3 stage for treating GBM brain cancer and their other product is DCVax-Direct for treating various inoperable cancers, it’s at Phase 1 in the clinic.

NWBO’s approach to treating cancer is unique and very exciting.
NWBO claims their dendritic cell based cancer vaccine is able to boost and direct the immune system to attack ALL cancer cell antigens. Cancers are known to have a large variety of antigens so it makes sense that attacking all of them is your best chance of defeating the cancer. Current immunotherapy treatments are mainly designed to attack a single cancer antigen so it is not surprising current treatments are having limited successes with fighting cancers. Monoclonal antibodies, checkpoint modulators and CAR-T cells are all examples of single agents that are currently used in treatments.

NWBO’s approach at treating cancer can theoretically be used to treat ALL types of solid tumors (operable and inoperable). So the potential market size for their treatments is huge. We have over 14M total cancer suffers in the US.

The market valuation of NWBO is around 420M, up to 5 times smaller than other small biotechs that have excited Wall Street. KITE, JUNO, BLUE are examples of companies enjoying much higher valuations, they have CAR-T cell treatments under development (single agent antigen treatments). NWBO should be valued at least at these same levels, especially considering their vaccines have a seemingly superior mechanism of action (attacking ALL cancer antigens instead of just one) and their vaccine is also showing an excellent and superior safety profile.

NWBO’s vaccines are also much further along in their clinical development. DCVax-L at Phase 3 and DCVax-Direct will soon to move to Phase 2 in the clinic. JUNO and KITE treatments are just entering the clinical stages.

Results from clinical trials of NWBO’s vaccines are very promising and probably not well understood by investors.
DCVax-L at Phase 2 showed excellent efficiency bettering the standard of care by up to 3 times in patient Overall Survival (OS), from 14.6 months to 36 months.
DCVax-L is also running a Phase3 “Compassionate” trail for patients that were TOO SICK and not eligible for their main Phase3 trial. Data is available from this trial of 55 patients. Median OS is about two times longer for DCVax-L than the SOC, 18 months versus 8.5 months. These results are a big positive for DCVax-L considering the patients being treated have tumors that are further advanced (and larger) than those patients being tested in their main Phase 3 trial. The vaccine also showed an excellent safety profile with fevers being reported after the vaccine injection which was treated with Tylenol.
 

Based on the data presented so far I suggest there is a high likelihood of success for this vaccine. NBWO’s relatively low valuation suggests the market does not understand the data presented so far. DCVax-L Phase 3 results due in early 2016.

The German FDA equivalent is impressed enough with early DCVax-L clinical results to approve it for compassionate use in German hospitals, even before Phase 3 results are out.
UK’s medical board is also considering early approval of DCVax-L. I suggest these medical bodies have a better handle on what these early DCVax-L trial results mean.   

NWBO has suffered from negative media coverage particularly from thestreet.com Adam Feuerstein.

I think this negative publicity has unfairly held back NWBO’s stock price. The main motivation for attacking NWBO may be its CEOs past where she was a vice-president at now infamous Enron.

As NWBO continues to progress its vaccines and show more positive data, as I expect, the media and investors will get behind and support NWBO because they truly offer a new and exciting hope for all cancer suffers.

2015 ASX Stockmarket Game1 - my tips

Game1 begins on the 26th of Feb and runs till the 10th of June, 15 weeks.

Hopefully this year's competition will be better and fairer than last years.
This comp had a trade execution flaw that allowed competitors to make unrealistic trades.
Becky, the winner of the last game, achieved a portfolio return of over 250%. Her return was completely unrealistic considering we were trading in a negative market and no single stocks returned anywhere near 250%.

The game organizers tell me they have fixed the flaw and the game will be now more realistic. Time will tell!
   
I will be constructing my fund with performance stocks. Performance stocks, with strong positive trends, tend to do well when the general market is buoyant. The ASX Index is off to strong start in 2015, up 14% since December's low, check the chart below:

TG Therapeutics (TGTX) trading at a low?!



TGTX is having a rough start in 2015. The stock is down over 22% from its highs at the beginning of this year.

However I am holding my position because I cannot find a good reason for this price-drop and I am not convinced it has broken its positive price-movement trend. 

TGTX’s current price is running at about its 75-day SMA (simple moving average) where it’s been several other times over the past 9 months so its price may just be trading at a low and presenting a good buying opportunity.
 
If TGTX's price moves beneath its SMA (75) this may signal a change to negative for its market sentiment and its price movement. In that case I will look to adjust my holding for the short-term.



The stock’s medium and longer term prospects still look great.

They kicked-off enrollments in their first Phase 3 clinical trial and are on track to deliver a bunch of clinical trial updates this year.
